Přehled We are seeking a highly motivated PhD student to join an interdisciplinary research project focused on Midichloria, a unique intracellular symbiont capable of invading mitochondria of oocytes of several tick species, e.g. Ixodes ricinus. Despite its exceptional biology, the mechanisms underlying mitochondrial specificity of Midichloria remain unknown. This project aims to identify metabolic and functional features that render mitochondria susceptible to bacterial invasion, addressing a fundamental question in host–symbiont interactions. The PhD student will investigate whether mitochondrial invasion is associated with distinct mitochondrial states reflecting altered metabolic activity or functional imbalance. Using comparative biological models, the project will examine mitochondrial physiology, stress-related and redox-associated processes in relation to symbiont localization, integrating imaging-based approaches with targeted molecular and biochemical analyses. In the later phase, the project will explore whether Midichloria can retain its mitochondrial targeting capacity beyond its native host environment, testing the robustness and specificity of this interaction across biological systems. Overall, this research will reveal general principles of mitochondrial recognition by intracellular bacteria and provide a conceptual framework for exploiting symbiotic interactions to modulate mitochondrial states.
